Today, it is commonly accepted that C. angulata and C. gigas are a single species according to morphological, genetic and experimental hybridization data. Following the viral disease that affected C. angulata and the subsequent expansion of C. gigas in Europe, it was of interest to examine the geographical distribution of both taxa in Europe. We studied the genetic composition of seventeen populations of cupped oysters, sampled in France, Spain, Portugal, Morocco and Italy, using a diagnostic marker, the mitochondrial Cytochrome Oxydase I gene. Results showed two distinct geographical zones. All the populations located in the northern part of the European sampled zone were mainly composed of C. gigas haplotypes whereas populations from southern Europe and... |

Exon Primed Intron Crossing (EPIC) markers providemolecular tools that are susceptible to be variable within specieswhile remaining amplifiable by PCR using potentially universal primers. In this studywe tested the possibility of obtaining PCR products from 50 EPIC markers on 23 species belonging to seven different phyla (Porifera, Cnidaria, Arthropoda, Nematoda, Mollusca, Annelida, Echinodermata) using 70 new primer pairs. A previous study had identified and tested those loci in a dozen species, including another phylum, Urochordata (Chenuil et al., 2010). Results were contrasted among species. The best results were achieved with the oyster (Mollusca) where 28 loci provided amplicons susceptible to contain an intron according to their size. This was... |

Trente... |

Widespread use of the herbicide atrazine has incited much research on its toxicity in aquatic systems, where it is routinely detected due to runoff from cultivated fields. Moreover, the determination of the genotoxic effect of such pollutants in the marine environment has become a major requirement for ecosystem protection. In the Pacific oyster, Crassostrea gigas, hypodiploid aneuploid cells have regularly been reported. There is a negative correlation between this phenomenon and growth, as well as evidence for a genetic basis. A positive relationship between atrazine and aneuploidy has previously been demonstrated in C. gigas adults and juveniles. To evaluate the persistence of this impact, our study examined the offspring of the same adult population... |

Despite the economic importance of oysters due to the high aquaculture production of several species, the current knowledge of oyster phylogeny and systematics is still fragmentary. In Europe, Ostrea edulis, the European flat oyster, and Ostreola stentina, the Provence oyster or dwarf oyster, are both present along the European and African, Atlantic and Mediterranean, coasts. In order to document the relationship not only between O. stentina and O. edulis, but also with the other Ostrea and Ostreola species, we performed a sequence analysis of the 16S mitochondrial fragment (16S rDNA: the large subunit rRNA-coding gene) and the COI fragment (COI: cytochrome oxidase subunit I). Oysters were sampled from populations in Portugal (two populations), Tunisia... |

Mangrove oysters include various species that are also present on different coasts. In the southern part of the Atlantic Ocean, Crassostrea rhizophorae has been observed along the southern coast of America and Crassostrea gasar along the western coast of Africa. Fifteen populations of these oysters were sampled from these two geographic areas. Their polymorphism was studied on the 16S mitochondrial fragment both by par sequencing and RFLP analysis. Some samples' caryotype were also analysed. Two haplotypes were identified: C. gasar was found in Africa, but also for the first time in South America, whereas C. rhizophorae was only found on the coasts of South America. |

Patterns of mitochondrial DNA (mtDNA) variation were used to analyse the population genetic structure of southwestern Indian Ocean green turtle (Chelonia mydas) populations. Analysis of sequence variation over 396 bp of the mtDNA control region revealed seven haplotypes among 288 individuals from 10 nesting sites in the Southwest Indian Ocean. This is the first time that Atlantic Ocean haplotypes have been recorded among any Indo-Pacific nesting populations. Previous studies indicated that the Cape of Good Hope was a major biogeographical barrier between the Atlantic and Indian Oceans because evidence for gene flow in the last 1.5 million years has yet to emerge. This study, by sampling localities adjacent to this barrier, demonstrates that recent gene... |

Nowadays, PCR-based DNA markers allow the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European flat oyster (Ostrea edulis) populations has aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Results were congruent between allozymes, microsatellites and mitochondrial DNA and support an isolation by distance model. High variability was observed in the breadth of diversity between populations using a mitochondrial marker, this highlights the small effective population sizes in some locations. A 10-fold quantitative difference was observed in Fst... |

Crassostrea gigas originated from the Pacific coast of Asia, but was introduced into several European countries in the early 1970s. Natural populations have now spread across the length of the western seaboard of Europe. To elucidate the demographic and selective processes at play during this rapid expansion, genome-scan analysis was performed on different populations. High diversities and low differentiation were observed overall, but significant genetic differentiation was found among newly established populations and between the newly established northern group and a nearly panmictic group composed of southern European populations and a population from Japan. Loss of genetic diversity was also seen in the north, likely caused by founder events during... |

P>Summer mortality is a phenomenon severely affecting the aquaculture production of the Pacific oyster (Crassostrea gigas). Although its causal factors are complex, resistance to mortality has been described as a highly heritable trait, and several pathogens including the virus Ostreid Herpes virus type 1 (OsHV-1) have been associated with this phenomenon. A QTL analysis for survival of summer mortality and OsHV-1 load, estimated using real-time PCR, was performed using five F-2 full-sib families resulting from a divergent selection experiment for resistance to summer mortality. A consensus linkage map was built using 29 SNPs and 51 microsatellite markers. Five significant QTL were identified and assigned to linkage groups V, VI, VII and IX. Analysis of... |

A breeding programme has been undertaken by Ifremer to produce selected Ostrea edulis families which are resistant to Bonamia ostreae, a parasite which is the most significant constraint on aquaculture of this oyster. Using such families, we aimed to identify Quantitative Trait Loci (QTLs) of resistance/susceptibility to bonamiosis. We used an F2 family derived from a bi-parental cross between a wild oyster and an individual from one of the selected oyster families. A total of 550 F2 individuals from the family cohabited with wild oysters that were injected with the parasite. Mortality was checked daily over six months and heart smears were used on moribund or dying oysters to detect the presence and level of infection of the parasite. Ninety two F2... |

The Pacific oyster, Crassostrea gigas, was introduced to Europe for aquaculture purposes, and has had a rapid and unforeseen northward expansion in northern Europe. The recent dramatic increase in number of C. gigas populations along the species' northern distribution limit has questioned the efficiency of Skagerrak as a dispersal barrier for transport and survival of larvae. We investigated the genetic connectivity and possible spreading patterns between Pacific oyster populations on the southern Norwegian coast (4 localities) and Swedish and Danish populations by means of DNA microsatellite analysis of adult oysters, and by simulating larvae drift. In the simulations we used a 3D oceanographic model to explore the influence of recent climate change... |

Rationale for EST and genome sequencing of the Pacific oyster: its membership of the Lophotrochozoa, an little studied branch of the Eukaryotes, its worldwide aquacultural importance (4.2 million metric tons, worth 3.5 billion US dollars), its high fecundity, with concomitantly high DNA polymorphism, its use as a biosensor of coastal pollution. In 2004, an international community of biologists, the Oyster Genome Consortium (OGC) led by D. Hedgecock (University of Southern California) and uniting 70 participants from 10 countries, first presented the Pacific oyster Crassostrea gigas as a genome-sequencing candidate to Joint Genome Institute (JGI, USA) (Hedgecock et al., 2005). Besides this project (that remains to be accepted), several EST... |
